    Effect of ultrasound intensification on the supercritical fluid extraction of phytochemicals from Agave salmiana bagasse

    [EN] The aim of this work was to evaluate the effect of ultrasound on supercritical fluid extraction for the recovery of antioxidants and saponins from agave bagasse as a green extraction technique. When a mass load of 0.086 g/cm(3) was used, ultrasound effect was not observed, due to sample swelling and compaction within the cell. For 0.043 g/cm(3), the intensification effect of ultrasound was significant (p < 0.05) and its magnitude depended on the transducer geometry. For a multiplate transducer geometry, antioxidant capacity increased from 12.18 +/- 1.01 to 20.91 +/- 1.66 mu mol TE/g; and saponins from 19.05 +/- 1.67 to 61.59 +/- 1.99 mu g/g, when ultrasound was applied. Although the amount of bioactives extracted is low, the use of a multiplate transducer design was able to intensify the supercritical fluid extraction of phytochemicals from agave bagasse. Se encuentra una relación estrecha entre modernidad empresarial y FPE. Utilizando funciones mincerianas modificadas se encuentra que la rentabilidad de la FPE aumenta con el nivel de educación formal y el tamaño de las empresas

    Variación latitudinal del ciclo reproductivo de dos bivalvos con orígenes biogeográficos contrastantes a lo largo del ecosistema de afloramiento de la Corriente de Humboldt

    Argopecten purpuratus (Lamarck, 1819) and Mesodesma donacium (Lamarck, 1818) are bivalves that inhabit the Humboldt Current Upwelling Ecosystem. They have contrasting biogeographical origins, suggesting that their responses to exogenous factors should differ. Using circular statistics, we examine synchrony/asynchrony in the reproductive cycle between populations of each species. The results indicate that there is reproductive asynchrony in both species along their distributional range. However, there was synchrony for A. purpuratus in several location-pairs, including Paita-Chimbote, Chimbote-Callao, Callao-Pisco and Pisco-Antofagasta. For M. donacium, there were only two synchronic groups: Camaná-Capellanía-Mehuín and Hornitos-Peñuelas-Longotoma-La Ligua-Cucao-Quilanlar. A. purpuratus showed gametogenenic activity throughout the year. In contrast, M. donacium showed strong seasonality, with gametogenesis in winter and spawning in spring/summer. In conclusion, the patterns observed for these sympatric species suggest that on a large scale the reproductive cycles follow the expected patterns for the contrasting biogeographic origin of each species, so it could be argued that they are modulated by endogenous factors. However, at a local scale, the reproductive cycles of these species show variation, likely determined by local oceanographic or hydrographic processes.Argopecten purpuratus (Lamarck, 1819) y Mesodesma donacium (Lamarck, 1818) son moluscos bivalvos endémicos del sistema de afloramiento de la Corriente de Humboldt pero tienen orígenes biogeográficos opuestos. Por tanto, se esperaría que los factores exógenos afectaran sus ritmos biológicos diferencialmente, entre ellos sus ciclos reproductivos. Mediante análisis de distribución circular se evaluó el sincronismo/asincronismo en ciclos reproductivos entre poblaciones de cada especie. Los resultados indican que existe asincronía reproductiva en ambas especies a lo largo de su distribución. Sin embargo para A. purpuratus se encontró varios pares de localidades sincrónicas: Paita-Chimbote; Chimbote-Callao; Callao-Pisco y Pisco-Antofagasta, y para M. donacium se dieron dos grupos sincrónicos, el primero entre las poblaciones de Camaná, Capellanía y Mehuín; y el segundo grupo entre Hornitos, Peñuelas, Longotoma, La Ligua, Cucao y Quilanlar. A. purpuratus presentó actividad gametogénica continua. En contraste, M. donacium presentó una marcada estacionalidad, con gametogénesis durante invierno y desoves durante primavera y/o verano. En conclusión, los patrones observados para ambas especies simpátricas sugieren que, a gran escala el ciclo reproductivo sigue los patrones esperados para el origen biogeográfico contrastante de cada especie, por lo tanto se podría argumentar que es modulada por factores endógenos. Sin embargo, a escala local el ciclo reproductivo de ambas especies muestra variaciones, aparentemente influenciados por los factores oceanográficos o hidrográficos locales

    Control of Thickness of PEDOT Electrodeposits on Glass/ITO Electrodes from Organic Solutions and its Use as Anode in Organic Solar Cells

    AbstractPoly-ethylendioxythiophene (PEDOT) was electropolymerized from the monomer EDOT in acetonitrile (ACN) containing Bu4N+ClO4-, BF4- or PF6- ions as supporting electrolyte. The electrode used was transparent electrodes (Glass/ITO) in order to generate the anode of an organic solar cell (OSC). Potentiodynamic and potentiostatic electropolymerization techniques were used to make the conducting polymer deposits (E-PEDOT), which were obtained as a thin film onto the ITO surface. It was possible to control the thickness of the electrodeposited films in the range of 15 to 200nm measured by AFM. With the thinner films (until 100nm), it was observed that its absorbance at 700nm was linearly dependent with their thickness and it was possible to obtain an equation that was used to measure the films thickness of future experiments. The E-PEDOT films were successfully used for constructing OSC's and the efficiency values found were equivalent or slightly superior to those found with the classical PEDOT:PSS anode

    Synthesis of Silver nanoparticles (AgNPs) with Antibacterial Activity

    The synthesis of nanomaterials is currently one of the most active in nanoscience branches; especially those help improve the human quality life. Silver nanoparticles (AgNPs) are an example of this as it is known to have inhibitory and bactericidal effects. In this work, we report the synthesis of silver nanoparticles by chemical reduction method of silver nitrate (AgNO3) from aqueous solution, using a mix of polivinyl pyrrolidone (PVP) - Aloe Vera as reducing agent and for stabilization and control of particle size. Silver nanoparticles obtained were characterized by Scanning Electron Microscopy (SEM), UV-visible spectroscopy and measurements using Zetasizer Nano ZS were applied to size estimation. The existence of surface plasmon resonance peak at λmax ∼ 420 nm is evidence of silver nanoparticles formation. It was possible to standardize an appropriate protocol for the evaluation of bactericidal activity of the nanoparticles, for mesophilic microorganisms. Bactericidal activity above 90% against these kinds of bacteria was demonstrated. © Published under licence by IOP Publishing Ltd

    Evaluación de técnicas de tinción para la observación de bandas de crecimiento en vértebras de elasmobranquios tropicales

    The aim of this study was to assess the suitability of different vertebrae staining techniques for the visualization and counting of growth bands in tropical species of batoids (Narcine leoparda, Urotrygon aspidura, Hypanus longus, Potamotrygon magdalenae) and sharks (Alopias pelagicus, Carcharhinus falciformis, Sphyrna lewini, Sphyrna corona and Mustelus lunulatus). Different cutting thicknesses and staining protocols were tested, analysing the precision and bias of each combination to identify the most accurate technique for estimating age. Vertebral sections of 0.4 mm were more suitable for batoids, except for Narcine leoparda; for this species and for all the shark species assessed, sections of 0.5 mm are recommended. Different combinations of stain and exposure time were required to achieve the best visualizations of vertebral growth band pair for the shark and ray species. Intraspecific variation occurred among vertebrae size of batoids. Our results confirm the importance of defining a suitable species-specific protocol for sectioning and staining hard structures before carrying out an age and growth study to improve the reliability of the age estimates.El objetivo de este estudio fue evaluar la efectividad de diferentes técnicas de tinción de vértebras en la visualización y el conteo de bandas de crecimiento en especies tropicales de batoideos (Narcine leoparda, Urotrygon aspidura, Hypanus longus, Potamotrygon magdalenae) y tiburones (Alopias pelagicus, Carcharhinus falciformis, Sphyrna lewini, Sphyrna corona y Mustelus lunulatus). Se probaron diferentes espesores de corte y protocolos de tinción, analizando la precisión y el sesgo de cada combinación para identificar la técnica más precisa para estimar la edad. Las secciones vertebrales de 0,4 mm fueron más adecuadas para batoideos, excepto para Narcine leoparda; para esta especie y para todas las especies de tiburones evaluadas, se recomiendan secciones de 0,5 mm. Se identificaron diferentes combinaciones de tinción y tiempo de exposición para lograr las mejores visualizaciones de las bandas de crecimiento vertebral en las especies de tiburones y rayas. En los batoideos se identificó variación intraespecífica de acuerdo con el tamaño de las vértebras. Nuestros resultados confirman la importancia de definir un protocolo especie-específico adecuado para cortar y teñir las estructuras duras antes de realizar un estudio de edad y crecimiento y así incrementar la confiabilidad de las estimaciones de edad

    Notch/Wnt Cross-Signalling Regulates Stemness Of Dental Pulp Stem Cells Through Expression Of Neural Crest And Core Pluripotency Factors

    Dental pulp stem cells (DPSCs) from adult teeth express neural crest (NC) markers together with core transcriptional factors associated with stem cell pluripotency, such as Oct4a, Sox2, c-Myc, Rex1, Stella/Dppa3, Ssea1/Fut4, Lin28 and Nanog. The possibility to boost the natural stemness features of DPSCs by mild methods, that do not involve gene and/or chromatin modification or gene transfection, is highly desirable for cell therapy. Canonical Wnt and Notch are two highly conserved developmental signalling pathways that are involved in NC emergence and stem cell self-renewal. We determined that both pathways coordinate to regulate the expression of core pluripotency and NC factors in DPSCs. Pharmacological inhibition of the Notch pathway for 48 h, by the gamma-secretase inhibitor N-[N-(3,5-Difluorophenacetyl)-L-alanyl]-S-phenylglycine t-butyl ester (DAPT), abolished the expression of NC and core factors. In addition, it induced a silencing of the canonical Wnt signalling and a clear reduction in the stemness potential of DPSCs, as shown by a reduced ability to generate mature, fully differentiated osteoblasts and adipocytes. Conversely, pharmacological activation of the Wnt pathway for 48 h, by either the glycogen synthase kinase 3 beta (GSK3-beta) inhibitor 6-bromoindirubin-3'-oxime (BIO) or the human recombinant protein Wnt-3a, not only largely increased the expression of NC and core factors, but also increased the efficiency of DPSCs to differentiate into mature osteoblasts and adipocytes. These results showed that a short preconditioning activation of Wnt/Notch signalling by small molecules and/or recombinant proteins enhanced the stemness and potency of DPSCs in culture, which could be useful for optimising the therapeutic use of these and other tissue-specific stem cells.Technical and human support provided by the analytical microscopy service of SGIKER (UPV/EHU, MINECO, GV/EJ, ERDF and ESF) is gratefully acknowledged.
